In the Neuro Probe AA10 chemotaxis chamber, chemoattractants added to the lower wells act on cells in a suspension added to the upper wells. A filter separates the upper and lower wells. After incubation the migrated adherent cells on the filter are stained and counted. In comparison with the more commonly used AP48 chamber, the AA10 has larger volume upper and lower wells and a larger exposed filter area.
The AA10 chamber consists of precision-machined acrylic top and bottom plates, assembly hardware, and a silicone gasket. The plates have ten wells in two offset rows of five wells each. Each well in the bottom plate is 8mm in diameter, holds 400µL, and has a rounded base. Corresponding holes in the top plate form the upper wells when the chamber is assembled. These top wells have a volume of 285µL. The exposed filter area is 50mm2. Also available by special order is the AA10-150, with a lower well volume of 150µL rather than 400µL. A single 25 x 80mm piece of filter membrane is placed between the top and bottom plates, and a gasket is positioned over the filter to create a seal. Narrow raised rims around mating surfaces of top and bottom wells enhance the seal.
Before the migrated cells can be stained and counted, the nonmigrated cells must be wiped off the top side of the filter. An accessory pack (Stock # P48AP) includes instruments for this purpose.
The AA10 is designed for use with polycarbonate filters, although cellulose nitrate filters may also be used.
Accompanying each chamber is an AA10 protocol for its use and care. Filters and accessories for the AA10 are the same as used with the AP48.
